REGENERATIVE PROPERTIES OF GRAPE CUTTINGS UNDER THE INFLUENCE OF TREATMENT WITH IAA
06.00.00 Agricultural sciences
Description
The article presents the research results of eight grapes rhizogenesis of different genetic origin of the cuttings after treatment with 0.01% solution of IAA for 24 h. It was found, that the activity of regenerative processes in grape cuttings depends on the varietal characteristics, storage conditions and the activation of IAA. It leads to disruption of existing natural correlations, including suppressing the longitudinal polarity. Root-growing ability of cuttings is a genetically determined feature: the effect of IAA did not cause the stability of the positive reaction; it is determined by genotype and depends on the quality and physiological condition of the cuttings

Description
Effects of different levels of vitamins (thiamine, pyridoxine and nicotinic acid) and macro-elements (MS, NN and PG) on development of globular, heart- and torpedo-stage embryos and plantlets with green cotyledons and on shoot production from them were studied in grapevine interspecific hybrids: cvs. ‘Bianca’, ‘Podarok Magaracha’ and ‘Intervitis Magaracha’. Pro-embryogenic calli were derived from petiole explants on solid full-strength NN medium supplemented with different levels of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzyladenine (BA) for each cultivar and used to initiate cell suspensions in liquid NN medium supplemented with 1 mg L-1 2,4-D and 0.2 mg L-1 BA. The following formulations of media were optimal for different stages of somatic embryogenesis and for plantlets with green cotyledons development: liquid NN medium supplemented with 0.5 mg L-1 BA for globular embryo formation; liquid HTE medium (with PG macro-elements, thiamine and pyridoxine at 5 mg L-1 each and 0.5 mg L-1 nicotinic acid): supplemented with 0.2 mg L-1 BA for heart-stage embryo development, supplemented with 0.1 mg L-1 indole-3-acetic acid (IAA) and 30 mg L-1 sodium humate for torpedo-stage embryo development and supplemented with 0.5 mg L-1 gibberellic acid (GA3) for the growth of plantlets with green cotyledons. Solid MS medium modified by the addition of nicotinic acid, pyridoxine and p-aminobenzoic acid (PAB) at 5 mg L-1 each, 0.5 mg L-1 thiamine and 0.5 mg L-1 BA was best efficient for shoot production from plantlets. Inflorescence formation in regenerated plants of cv. ‘Podarok Magaracha’ grown on solid hormone-free PG medium was induced by addition of nicotinic acid, pyridoxine and PAB at 5 mg L-1 each, 0.5 mg L-1 thiamine and 0.2 mg L-1 BA into liquid modified HTE medium for growth of plantlets with green cotyledons and into solid modified MS medium for shoot production from them.

Description
The article discusses the use of automatic systemic-cognitive analysis (ASC-analysis), its mathematical model is a system of information theory and software tools – an intellectual system called "Eidos" for the solution of some problems of ampelography: 1) digitization of scanned images of the leaves and creation of their mathematical models; 2) the formation of mathematical models of specific leaves using the spreading of information theory; 3) the formation of models of generalized images of leaves of various sorts; 4) comparing an image of a specific leaf with a generalized image of the leaf of different varieties and finding a quantitative degree of similarity and differences between them, i.e. the identification of the varieties on the leaf; 5) quantification of the similarities and differences of the varieties, i.e. cluster-constructive analysis of generalized images of the leaves of different varieties. We propose a new approach to digitizing images of leaves, based on using the polar coordinate system, the center of gravity of the image and its external contour. Before scanning images we may use transformation to standardize the position of the still images, their sizes and rotation angle. Therefore, the results of digitization and ASC-analysis of the images might be invariant (independent) relatively to their position, size and rotation. The specific shape of the contour of the leaf is regarded as noise information on the variety, including information about the true shape of the leaf of the class (clean signal) and noise, which distort this true form, originating in a random environment. Software tools of ASC-analysis – intellectual "Eidos" system ensures noise reduction and the selection of the signal about the true shape of the leaf of each variety on the basis of a number of noisy concrete examples of the leaves of this variety. This creates a one way form of a leaf of each class, free from their concrete implementations, i.e., the "Eidos" of these images (in the sense of Plato) is a prototype or archetype (in the Jungian sense) of the images

CROSSABILITY OF CRIMEAN INDIGENOUS GRAPE VARIETIES WITH FORMS OF VARIOUS ORIGIN
06.00.00 Agricultural sciences
Description
Progress of breeding for resistance to diseases, pests and low temperatures is provided by cross-species hybridization. Institute of Vine and Wine "Magarach" is one of the leading centers of selection of grapes in the world. The breeding program of deducing grapes at the institute is based on the study and attracting the world's gene pool, including and industrial of the Russian Federation assortment of grapes. Analysis assortment shows that the share of productive, highquality varieties with group resistance to diseases, pests and extreme environmental factors is very small. The main method of selection of grapes of a new generation who have genetically determined traits earliness, resistance to biotic and abiotic factors is crossing complex interspecific hybrids and forms of their involvement with each other and with the forms of species Vitis vinifera, belonging to different ecogeographical groups, saturating crossing between hybrid forms with complex features, the appropriate selection of reference. For vigorous offspring is most effective as a parent to use a local variety of forms of Crohn and father - interspecific cultivar Alminsky

STRUCTURAL AND PHYLOGENETIC ANALYSIS OF VVMYBA1, VVMYBA2 AND VVMYBA3 GRAPEVINE GENES
Description
The article presents the structural and phylogenetic analysis of VIT_02s0033g00410, VIT_02s0033g00390 and VIT_02s0033g00450 genes of the grapevine genome and the closely related orthologous gene MYB114 of the Arabidopsis genome. These genes are responsible for the biosynthesis of anthocyanin in the organs of model plants and are of interest not only for practical production and breeding, but also for fundamental research. These genes were analyzed for GCcomposition of nucleotides, the presence of cisregulatory elements and promoter regions. DNA and protein sequences were aligned to look for similar elements, which allowed further analysis of the ultraconservative domains of four genes. Based on the results of search and identification of the conservative regions, a cluster tree was constructed, which made it possible to identify the separation of gene sidelines from, presumably, the main one. At the same time, the construction of consensus trees based on DNA and protein sequences revealed their absolute similarity. "The Minimal Evolution Tree" allowed calculating the approximate dates of the appearance of the mutations and the divergence times of the gene branches between each other. At the same time, the appearance of the Vitis genus and its separation from the Rosales was taken as the time first divergence point. In the end, homologous metabolic pathways were searched between grapevine and Arabidopsis, which revealed the presence of homologous proteins in the grape proteome. In this turn, it already confirms the existence of similar biosynthetic pathways and, as a consequence, interactions such as "DNA-protein" and "protein-protein"
