DEVELOPMENT OF MULTIPLEX SETS OF SSR MARKERS FOR GENOTYPING APRICOT VARIETIES (PRUNUS ARMENIACA L.)
Description
Genetic studies of apricot are the actual direction in the genetics of fruit crops. In this regard, the improvement of the collection of SSR markers for the genotyping of this culture is an objectively significant task. In a study for the 16 SSR-markers previously developed on almonds (PdUnchar2, PdSLD1, PdGMGT1, PdTrTFGT1, PdUnchar2, PdSLD1, PdGMGT1, PdTrTFGT1) and Siberian apricot (A3-72, A1-63, H2-22, A3- 7-1, H2-5, A1-7, A3-9, H2-45), approbation and evaluation of the prospects of using for genotyping Prunus armeniaca L. were performed. Approbation, performed on 3 varieties of different origin, revealed markers and their combinations optimal for their use. During the study, all tested DNA markers were grouped into multiplex sets, including 4 markers. This allows carrying out genotyping simultaneously on 4 loci in the formulation of one reaction. One marker (PdUnchar2) from the studied sample included in the multiplex set did not show amplification. Five markers gave a monomorphic product. The remaining 11 SSR markers allowed us to obtain polymorphic, cultivar-specific SSR fingerprints for all the studied cultivar. These multiplex sets are proposed for use in studying the genetic polymorphism of the species Prunus armeniaca L.

MODERN TECHNOLOGIES FOR GRAPE STORING

Description
The article gives a review of modern technologies of storage of grapes. The main tool for stabilizing the quality of grapes during transportation and storage is the use of sulfur dioxide (SO2), a gas that has properties that can inhibit non-enzymatic darkening, reduce the activity of enzymes, effectively control the development of Botrytis cinérea (a phytopathogen that causes gray rot) and acts as an antioxidant. Researchers pay particular attention to control the release rate of SO2, as it is important in the first stage storage to ensure supply of significant quantities of SO2 for a short period of time, which eliminates Botrytis spores that are present on the surface of berries and to stabilize the damages formed during transport and filling; the second step is to ensure a steady supply of the minimum quantity of the SO2, to ensure the control of microbiological spoilage. For a long storage time. To preserve the quality indicators of grapes during storage, it is possible to use a modified gas environment created in various ways, including through special types of packaging; forced saturation of the environment with gases such as oxygen, ozone or carbon dioxide. At the same time, to enhance the effect can additionally apply fungicides, antifungal ear Anti-mold®, etc. There are known technologies for complex processing of table grapes to combat post-harvest microbiological spoilage, which provide for the first stage of treatment with ozone or sulfur dioxide followed by spray treatment with Muscodor albus. An alternative to the use of SO2 is the use of essential oils and hypobaric treatment methods that ensure minimal environmental impact. Positive results during storage are provided by the use of medium-range ultraviolet radiation (UV-B) or short-wave radiation (UV-C), which positively affect the transcription of biosynthetic genes, providing an increase in the content of phenolic compounds and antioxidant activity of grapes, without affecting the content of dry soluble substances, the value of total titrated acidity and pH during storage. GRAPE VARIETIES OF THE ANAPA AMPELOGRAPHIC COLLECTION AND THEIR USE IN BREEDING WORK

Description
In the world, huge work is being done to create global information banks of plant genetic resources. The need for conservation and rational use of genetic resources in modern conditions has become very relevant. Genetic collections are the basis for selection work: creating new varieties, replenishing the assortment with new, classic, introduced and native varieties, adapted to the natural (soil and climatic) conditions of the cultivation sites. In the Anapa ampelographic collection, 4911 varieties of various ecogeographical and genetic origin are represented, the use of which provides great opportunities for creating and breeding new varieties, their state testing and zoning. In the gene pool of the ampelographic collection, most varieties of V. vinifera L. grapes are represented by local honeycombs of different regions of the grape culture, and more than one fourth of varieties of V. vinifera L. are obtained from intraspecific crossings. Despite the value of local varieties of grapes, they do not always meet all the requirements of production. Therefore, to improve the local assortment, it is necessary to carry out selection - breeding new varieties of grapes. This article presents the long-term results of the work on the selection of varieties of table and technical directions in the Anapa ampelographic collection used for breeding. Also, the role of the genetic collection in the creation of new generation varieties by combinative selection is shown - as donors for creating new varieties of grapes. Dedicated varieties are the sources of valuable traits, which are given primary importance and are especially important for the viticulture of the Russian Federation

PHENOTYPING OF HIGH-QUALITY TECHNICAL GRAPES OF AZOSVIV SELECTION IN THE AMPELOGRAPHIC COLLECTION

Description
A variety determines the direction of use of grape products and plays a leading role in improving its quality. Preservation of genetic resources of grapes and their studying are very important for modern science. The studied material makes it possible to use the selected genetic characteristics for breeding new varieties of grapes, taking into account the requirements, and introducing them into production, as well as highlighting the most valuable varieties for transferring them to the state variety testing. Among a wide variety of grape varieties, in the ampelographic collection there are varieties of AZOSVIV selection of the technical direction: Krasnostop AZOS and Dostoiniy, for which complete phenotyping was carried out. The phenological and agrobiological information on the grape varieties studied for 2015- 2017 is collected. The presented grades of grapes of the technical direction are characterized by high quality of production. The wines from the varieties Krasnostop AZOS and Dostoiniy are competitive, characterized with high consumer properties; they enjoy the well-deserved fame and were nominees of exhibitions and holders of diplomas repeatedly. Based on the results of microsatellite profiling, the varieties showed a sort-specific general combination of alleles. At the same time, in each locus there is one identical allele, which corresponds to the fact that the varieties Krasnostop AZOS and Dostoiniy have one common parent (Phylloxer-resistant Gemete). The received formulas of a variety (DNA-passports) in practice can be effectively applied to determine the cleanliness of planting stock and plantations of grapes, specification of the parent forms of the sample and in the disputed questions of the authorship of the variety.
