ISSUE OF DEVELOPMENT OF NONWEEDING TECHNOLOGY OF GROWING RICE SEEDS

DescriptionAn analysis of the state of contamination of rice crops with red-grained forms was carried out, as well as impurity was found both in the seed material and in the soil. It was shown that there is a need to switch to non-weeding seed growing technology when conducting elite and reproductive seed production. Various variants of soil cultivation in combating the red rice forms are considered, both in the main and in the pre-sowing treatment. It has been found that after harvesting rice on heavily foul areas, it is better to burn straw, and not to carry out autumn tillage of the soil, the seeds of red-grained forms die mostly when wintering on the soil surface. The most effective method of soil cultivation for controlling of red rice forms in seed crops has been identified, in which the yield is increased, and the contamination of crops and the resulting rice grain is significantly reduced. The work carried out will allow reducing manual labor costs in the production of seeds, improving their quality and assortment, speed up variety changing and variety updating

Most intensively, these processes are developed in meadowbog soils, confined to closed depressions of the plains plain. Meadow-black soils lying on elevated relief elements have more favorable physical, physicochemical and oxidation-reduction properties for cultivating rice and accompanying crops in crop rotation. DescriptionIn breeding programs, evaluation of initial material is made on all the stages of developing the variety. Great difficulties for the breeding process are made by absence of modern mechanisms for storing, retrieving, manipulating the relevant data, and using the information previously obtained for use in solving the set breeding task. In order to organize the management of breeding process for development of high-quality rice varieties, based on the urgency of problem of intellectual analysis of data, the Database "Rice Quality" was designed as a set of structured data - the rice quality characteristics identified in the stages of the process of developing variety belonging to the "Rice". In the domain model, the infologic model, a set of parameters for evaluating rice quality in terms of out-of-program components is defined. The program is written in the Microsoft Access database environment using built-in query building tools, forms, reports. The structure of the developed tables includes static and refreshed directories in which the necessary information is stored, the tables are combined into one-to-many relationships, while ensuring data integrity, cascading updates and deletion of fields

A MODIFIED PROTOCOL OF RNA ISOLATION FROM MATURE LEAVES OF GRAPES FOR RT-PCR
DescriptionIsolation of high-quality RNA from the tissues of perennial woody plants, including woody grape vines, is very difficult due to the high content of phenolic compounds, secondary metabolites and polysaccharides and the ribonuclease activity of destroyed tissues. Most of the existing methods require either large time or financial costs, or do not give reproducible results in the case of RNA extraction from mature grape tissues. The modified isolation protocol is based on a combination and modification of the known RNA extraction methods, taking into account the characteristics of mature grape tissues. Existing commercial kits for the isolation of RNA from plant tissues showed a low efficiency of RNA extraction from mature grape tissues, primarily associated with "varietal specificity". Reproducible results in the extraction of RNA showed CTAB-method, however, it has several significant drawbacks associated with the duration of the extraction and the complexity of the processing of an RNA preparation with a DNAase. The developed method is based on increasing the concentration of mercaptoethanol and polyvinylpyrrolidone in the extraction buffer, eliminating the stage of RNA selective precipitation via LiCl, and replacing it with deposition on a silica-based membrane (SiO2) followed by processing with DNA-ase. and increase the purity of the preparation of RNA from genomic DNA in comparison with the original method. INHERITANCE OF PRODUCTIVITY TRAITS IN RICE HYBRIDS
DescriptionGreat success in increasing rice production, and it has more than doubled, has occurred worldwide thanks to the use of modern high-yielding rice varieties developed by conventional breeding methods. In order to develop a new variety, it is important for the breeder to study the genetic and breeding value of various cultural and wild species - carriers of economically valuable traits, the identification of genes and groups of genes controlling the economically valuable traits, to study the patterns of their inheritance, to establish patterns of interaction between the genotype and the environment, explore the genetic and physiological-biochemical basis of the phenomenon of heterosis, improve methods of intraspecific, complex, stepwise and distant hybridization, to study a phenomenon of a sharp increase in the quantitative characteristics of crop plants, a more powerful growth of hybrids of the first generation compared with the parent individuals. Scientists of the past centuries, such as A.F. Wigman, W.Noden, V.Foke, and many others have been studying the phenomenon of hybrid power, which was first named “heterosis” in 1908 by J. Schell. At present, heterosis is understood as the phenomenon when hybrids of the first generation exceed their parental forms in terms of their biological values and economically valuable traits. The magnitude of heterosis of the first generation hybrids is determined by several methods in terms of percentages to the paternal, to the maternal form, to the average of both parents, and to a better or regionalized standard. DEVELOPMENT OF MULTIPLEX SETS OF SSR MARKERS FOR GENOTYPING APRICOT VARIETIES (PRUNUS ARMENIACA L.)
DescriptionGenetic studies of apricot are the actual direction in the genetics of fruit crops. In this regard, the improvement of the collection of SSR markers for the genotyping of this culture is an objectively significant task. In a study for the 16 SSR-markers previously developed on almonds (PdUnchar2, PdSLD1, PdGMGT1, PdTrTFGT1, PdUnchar2, PdSLD1, PdGMGT1, PdTrTFGT1) and Siberian apricot (A3-72, A1-63, H2-22, A3- 7-1, H2-5, A1-7, A3-9, H2-45), approbation and evaluation of the prospects of using for genotyping Prunus armeniaca L. were performed. Approbation, performed on 3 varieties of different origin, revealed markers and their combinations optimal for their use. During the study, all tested DNA markers were grouped into multiplex sets, including 4 markers. This allows carrying out genotyping simultaneously on 4 loci in the formulation of one reaction. One marker (PdUnchar2) from the studied sample included in the multiplex set did not show amplification. Five markers gave a monomorphic product. The remaining 11 SSR markers allowed us to obtain polymorphic, cultivar-specific SSR fingerprints for all the studied cultivar. These multiplex sets are proposed for use in studying the genetic polymorphism of the species Prunus armeniaca L.

CJSC Priazovskoye located in the Slavyansk district is in the area of the Priazovie marshes. Soil and climatic conditions make it possible to grow many heat-loving crops including rice. The farm specializes in the cultivation of rice and the production of meat and cattle dairy products. The rice system is divided into 8-crop rotations, where, in addition to rice, animal feed is grown. Livestock waste is used as organic fertilizer in the fields of crop rotation. In the field experiment with perennial grasses (alfalfa) as predecessor, the change in yield and seed quality of three rice varieties Rapan, Diamant and Flagman was studied. The rates of the applied fertilizers were: phosphorus 100 kg/ha, potassium 60 kg/ha and increasing rates of nitrogen from 30 to 120 kg/ha. It has been established that the optimal rates of nitrogen fertilizers after alfalfa are 90 kg/ha (a.i.) for Rapan and Diamant varieties and 60 kg/ha (a.i.) for Flagman. At the same time, the maximum grain yield was obtained for Rapan - 7.45 t/ha, Diamant - 8.27 t/ha, Flagman - 7.62 t/ha , and the best seeds were formed in terms of germination and emergence. Analysis of the crop structure of rice varieties showed that with increasing rates of nitrogen the productive tillering and grain mass per panicle increased. With the nitrogen rate higher than the optimum, the number of unfilled grains per panicle and the grain /straw ratio have increased. Based on this analysis, it can be considered that the number of unfilled grains per panicle and the grain /straw ratio can serve as indicators of the response of rice plants of these varieties to an excess of the optimal level of mineral nutritions

RICE QUANTITATIVE TRAITS CONTROLLING YIELD AND THEIR INHERITANCE
DescriptionRice is a crop with high yields guaranteed by nature itself. In recent years, huge changes have occurred in the domestic rice cultivation, but despite this, there are still a lot of unsolved relevant problems. One of such problems is the development of high yielding varieties. The success of breeding largely depends on the availability of genetically diverse starting material with a wide response to changing environmental conditions, its knowledge and proper selection; as well as the identification of new sources of valuable traits and properties, among ecologically remote agro-types that enrich the genetic potential of domestic varieties. Development of a high-yielding variety is greatly influenced by economically important traits: plant height, total and productive tillering, the number of spikelets and grains in the panicle, grain size, weight of 1000 grains, etc. In modern biological science of heredity, there is still the question of the inheritance of quantitative traits. The study of inheritance of quantitative traits constituting the main elements of the yield structure is one of the most important tasks, the solution of which is necessary to speed up the breeding process. CURRENT STATE AND EFFICIENCY OF BLACK LEACHED SOILS OF WESTERN CAUCASUS
DescriptionThe article studies current state of fertility and productivity of leached chernozem used in the 11-field grain-grass-row crop rotation. Under the conditions of a stationary field experiment, a soil section was laid, a morphological description of the soil was carried out, its magnetic susceptibility χ was measured, soil samples were selected from soil-genetic horizons, in which the particle size distribution, physical and chemical parameters were determined. Studies have shown that leached chernozem is characterized by an addition density of 1.30 g/cm3, a solid phase density of 2.67 g/cm3, a total porosity of 51.9 %, χ = 1,045 × 10-3 units SI, pHwat. 6.5 units, humus content 3.2 %, its stock in the layer A + B = 468.2 t/ha, the sum of the exchange Ca2+ and Mg2+ 42.8 mg.eq./100 g. The use of mineral fertilizers increases the productivity of leached chernozem: the yield of sunflower of 11-field grain-grass-row cultivated crop rotation exceeds the control without fertilizers by 1.02-1.62 t/ha

DescriptionThe article presents results of studies on the intraspecific structure of the population of blast causative agent using sets of tester varieties from International Rice Research Institute - IRRI. The differentiation of the fungus population on tester varieties is the most progressive, since it allows one to determine the virulence genotypes of races of pathogens and identify the resistance genes of rice plants effective for them. The work has studied the diversity of blast causative agent in released rice varieties by morphological and cultural traits. Currently, in the world, the resistance of rice plants to blast is becoming one of the important indicators of the competitiveness of varieties. The basis for the successful rice breeding for resistance to blast is the availability of source material with a fairly wide spectrum and a high level of resistance.
